Scientific Data
Detection of Human and Mouse SHIP by Western Blot. Western blot shows lysates of BaF3 mouse pro-B cell line and RL human non-Hodgkin's lymphoma B cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human/Mouse SHIP Antigen Affinity-purified Polyclonal Antibody (Catalog # AF10117) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (HAF017). A specific band was detected for SHIP at approximately 145 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Detection of SHIP by Immunoprecipitation. PMA-treated U-937 lysates were prepared and immunoprecipitation was performed using 2.0 µg of Goat Anti-Human/Mouse SHIP Antigen Affinity-purified Polyclonal Antibody (Catalog # AF10117) pre-coupled to Dynabeads protein G. Immunoprecipitated SHIP was detected with Mouse Anti-SHIP Antibody. The Ponceau stained transfer of the blot is shown. SM=4% starting material; UB=4% unbound fraction; IP=immunoprecipitate; HC=antibody heavy chain. Image, protocol and testing courtesy of YCharOS Inc. (ycharos.com).

Background: SHIP
SH2-containing inositol phosphatase (SHIP), also known as INPP5D, is a negative regulator of signal transduction in hematopoietic cells. Targeted disruption of SHIP in mice leads to a myeloproliferative disorder. Several laboratories have demonstrated the presence of multiple forms of SHIP, including 145 kDa, 135 kDa, and
C‑terminal truncated forms at 125 kDa and 110 kDa in some cell types.
