RSK1/RSK2 in Mouse Brain. RSK1/RSK2 was detected in perfusion fixed frozen sections of mouse brain (Purkinje cells in cerebellum) using 15 µg/mL Rabbit Anti-Human/Mouse/Rat Phospho-RSK1/RSK2 (RSK1 S221, RSK2 S227) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF892) overnight at 4 °C. Tissue was stained with the Anti-Rabbit HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS005) and counterstained with hematoxylin (blue). Detection of Human Phospho-RSK1 (S221) and RSK2 (S227) by Western Blot. Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line untreated (-) or treated (+) with 200 nM PMA for for the indicated times. PVDF membrane was probed with 0.5 µg/mL of Rabbit Anti-Human/Mouse/Rat Phospho-RSK1 (S221)/RSK2 (S227) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF892), followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Phospho-RSK1 (S221) and RSK2 (S227) at approximately 90 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: RSK1/RSK2

The 90 kDa ribosomal protein S6 kinases (RSK1-4) are a family of broadly expressed Ser/Thr kinases activated in response to mitogenic stimuli, including growth factors and tumor-promoting phorbol esters. RSKs are directly phosphorylated by ERK1 and ERK2, leading to RSK autophosphorylation and activation. Active RSKs appear to play a major role in transcriptional regulation by translocating to the nucleus and phosphorylating c-Fos and CREB.
