Applications

Recommended Concentration
Sample
Neutralization
Measured by its ability to neutralize CCL19/MIP‑3 beta -induced chemotaxis in the BaF3 mouse pro-B cell line transfected with human CCR7. The Neutralization Dose (ND50) is typically 0.4-2.4 μg/ml in the presence of 25 ng/mL Recombinant Rat CCL19/MIP‑3 beta.

Please Note: Optimal dilutions should be determined by each laboratory for each application. General Protocols are available in the Technical Information section on our website.

Chemotaxis Induced by CCL19/MIP‑3 beta and Neutralization by Rat CCL19/MIP‑3 beta Antibody. Recombinant Rat CCL19/MIP-3 beta (Catalog # 7800-M3) induces chemotaxis in the BaF3 mouse pro-B cell line transfected with human CCR7 in a dose-dependent manner (orange line). Chemotaxis elicited by Recombinant Rat CCL19/MIP-3 beta (25 ng/mL) is neutralized (green line) by increasing concentrations of Mouse Anti-Rat CCL19/MIP-3 beta Monoclonal Antibody (Catalog # MAB7800). The ND50 is typically 0.4-2.4 µg/mL.

Background: CCL19/MIP-3 beta

CCL19 (also known as Mip-3 beta, ELC, CK beta -11, Scya19 and Exodus-3) is a 9 kDa secreted member of the CC family of chemotactic cytokines (1-4). Cells known to express CCL19 are limited in number, and include activated monocytes (5), stromal cells in nodal T zones (6), CD8+ dendritic cells (DC) (6), vascular endothelial cells (7), visceral smooth muscle and mast cells (8), and thymic stromal cells (9).  Rat CCL19 is synthesized as a precursor that is 1108 amino acids (aa) in length. Mature rat CCL19 shares 72% and 89% aa sequence identity with human and mouse CCL19, respectively. CCL19 is known to bind to CCR7 (3, 4), CCLR2/CRAM (L-CCR in rodent) (10, 11) and CCX-CKR (12), with the last two receptors representing scavenger, or chemokine-sink receptors.
