Detection of Mouse Siglec‑3/CD33 by Western Blot. Western blot shows lysates of EL-4 mouse lymphoblast cell line. PVDF membrane was probed with 2 µg/mL of Rat Anti-Mouse Siglec-3/CD33 Monoclonal Antibody (Catalog # MAB2220) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for Siglec-3/CD33 at approximately 65 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Siglec-3/CD33

Siglecs (Sialic acid binding Ig-like Lectins) are I-type (Ig-type) cell-surface lectins that recognize sialic acid within specific glycan structures. Mouse Siglec-3, also known as CD33 and GP67, is expressed primarily on myeloid precursors in the bone marrow and on granulocytes in peripheral blood. Two isoforms have been reported for mouse Siglec-3 that display distinct cytoplasmic domains resulting from differential mRNA splicing.
