Scientific Data
Detection of Human Rictor by Western Blot. Western blot shows lysates of HEK293 human embryonic kidney cell line and HeLa human cervical epithelial carcinoma cell line. PVDF membrane was probed with 1 µg/mL of Human Rictor Monoclonal Antibody (Catalog # MAB4598) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for Rictor at approximately 200 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Rictor
Rictor (rapamycin-insensitive companion of mTOR) is a member of the mTORC2 complex, which regulates cell growth by integrating stress, energy, and nutrient signals with growth factor derived signals. Rictor contains no obvious catalytic motifs, but knockdown studies of mTOR or Rictor results in a loss actin polymerization and cell spreading. In vitro, the mTOR-Rictor complex phosphorylates Akt/PKB on Ser473, an essential step for full activation of Akt/PKB. Additionally, recent studies have shown that mTOR-Rictor regulates PKC alpha phosphorylation and PDGFR beta expression and signaling. Rictor has also been described as a protein substrate for Akt.
