Detection of Human Phospho-TOR (S2448) by Western Blot. Western blot shows lysates of MCF-7 human breast cancer cell line treated (+) with 1 µM Camptothecin (CPT) for 5 hours. PVDF membrane was treated (+) or untreated (-) with 300 U/mL CIP for 1 hour and probed with 1 µg/mL of Rat Anti-Human Phospho-TOR (S2448) Monoclonal Antibody (Catalog # MAB1665) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for Phospho-TOR (S2448) at approximately 280 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: TOR

The mammalian Target of Rapamycin (mTOR) is a member of the PI 3-kinase-related kinase (PIKK) family. mTOR is the protein target of rapamycin, an anti-rejection drug used in transplantation and promising anti-cancer agent. Recent evidence suggests that mTOR is phosphorylated at S2448 by the S6 protein kinase 1 (S6K1) (1, 2), rather than a direct phosphorylation by the Akt protein kinase. This S2448 phosphorylation activates mTOR to affect the downstream control of cell growth and survival.
