Detection of PD-L1/B7-H1 in MDA-MB-231 human breast adenocarcinoma cell line by Flow Cytometry. MDA-MB-231 human breast adenocarcinoma cell line was stained with Human Anti-Human PD-L1/B7-H1 (Research Grade Atezolizumab Biosimilar) Monoclonal Antibody (Catalog # MAB10348, filled histogram) or irrelevant antibody (open histogram) followed by APC-conjugated Anti-Human IgG Secondary Antibody (F0135). PD-1 binding to Human PD-L1/B7-H1-transfected HEK293 Human Cell Line is Blocked by Human PD-L1/B7-H1 Antibody. In a functional flow cytometry test, Recombinant Human PD-1 His-tagged protein (8986-PD, 500 ng/mL) binds to HEK293 human embryonic kidney cell line transfected with recombinant human PD-L1/B7-H1. (A) Binding is completely blocked by 2.5 µg/mL of Human Anti-Human PD-L1/B7-H1 (Research Grade Atezolizumab Biosimilar) Monoclonal Antibody (Catalog # MAB10348) but not by (B) Human IgG Isotype Control (1-001-A). Ligand binding was detected by staining cells with Anti-His tag APC-conjugated Monoclonal Antibody (IC050A).

  1. Does this Biosimilar antibody have the same sequence as the therapeutic antibody?

    • The Biosimilar antibody has the same variable region and constant region sequence as the therapeutic antibody, except for Catalog numbers MAB9677, MAB9575, MAB9577, MAB9589, and their conjugates. These four antibodies have the same variable region as the Biosimilar, but not the same constant region.

  2. Is the Fc portion of Human PD-L1/B7-H1 (Research Grade Atezolizumab Biosimilar) Antibody, Catalog # MAB10348, inert, similar to what has been reported with the therapeutic Atezolizumab?

    • We have not performed any testing in house to confirm that the Fc portion is inert, but the sequence of MAB10348 is the same as the therapeutic Atezolizumab.
