Detection of Human p15INK4b/CDKN2B by Western Blot. Western blot shows lysates of 293T human embryonic kidney cell line and HeLa human cervical epithelial carcinoma cell line. PVDF Membrane was probed with 1 µg/mL of Mouse Anti-Human p15INK4b/CDKN2B Monoclonal Antibody (Catalog # MAB6798) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for p15INK4b/CDKN2B at approximately 15 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
p15INK4b/CDKN2B in HeLa Human Cell Line. p15INK4b/CDKN2B was detected in immersion fixed HeLa human cervical epithelial carcinoma cell line using Mouse Anti-Human p15INK4b/CDKN2B Monoclonal Antibody (Catalog # MAB6798) at 3 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. Background: p15INK4b/CDKN2B
p15INK4B is an approximately 15 kDa member of the p16INK4 family. Human p15INK4B is 138 amino acids (aa) in length and contains four ankyrin repeats, a common structural sequence found in members of the INK4 family. The first 50 aa of p15INK4B share 44% sequence identity with p16INK4. Following this is an 81 aa region sharing 97% sequence identity, after which the two proteins diverge. Human p15INK4B shares 83% aa sequence identity with mouse and rat p15INK4B. Functionally, p15INK4B interacts strongly with CDK4 and CDK6 and is a potent inhibitor. p15INK4B can potentially induce cell cycle arrest by acting as an effector of TGF-beta.
