Detection of Human THAP11 by Western Blot. Western blot shows lysates of BG01V human embryonic stem cells. PVDF membrane was probed with 1 µg/mL of Human THAP11 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5727) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for THAP11 at approximately 45 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8.

Background: THAP11

THAP11, also known as Ronin, is a 36-44 kDa member of the thanatos-associated protein family. It is ubiquitously expressed and through its repression of c-Myc transcription, blocks cell proliferation. Human THAP11 is 314 amino acids (aa) in length. It contains a DNA-binding THAP zinc-finger domain (aa 5-81), an NLS (aa 98-104), a polyGln region (aa 104-146), an Ala-rich segment (aa 197-221) and a C-terminal coiled-coil region (aa 255-305). THAP11 is phosphorylated on Thr25. Over aa 226-314, human and mouse THAP11 are 100% identical in amino acid sequence.
