Detection of Human and Mouse Haptoglobin by Western Blot. Western blot shows human plasma. PVDF membrane was probed with 0.1 µg/mL of Goat Anti-Human/Mouse Haptoglobin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8465) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for Haptoglobin at approximately 45 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Detection of Human Haptoglobin by Simple WesternTM. Simple Western lane view shows human serum, loaded at 1:25000 dilution. A specific band was detected for Haptoglobin at approximately 60 kDa (as indicated) using 1 µg/mL of Goat Anti-Human/Mouse Haptoglobin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8465) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Human Haptoglobin ELISA Standard Curve. Recombinant Human Haptoglobin protein was serially diluted 2-fold and captured by Mouse Anti-Human Haptoglobin Monoclonal Antibody (Catalog # MAB8465) coated on a Clear Polystyrene Microplate (Catalog # DY990). Goat Anti-Human/Mouse Haptoglobin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8465) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (Catalog # DY998) followed by Substrate Solution (Catalog # DY999) and stopping the enzymatic reaction with Stop Solution (Catalog # DY994).

Background: Haptoglobin

Haptoglobin is a 406 aminoacids (aa) protein that in humans is encoded by the HP gene. This gene encodes a preproprotein that is processed into alpha (aa 19-160) and beta (aa 162-406) chains that combine as a tetramer to produce haptoglobin. Haptoglobin captures, and combines with free plasma hemoglobin to allow hepatic recycling of heme iron and to prevent kidney damage.
