Detection of EMR3 in Human Blood Monocytes by Flow Cytometry. Human peripheral blood monocytes were stained with Mouse Anti-Human CD14 APC-conjugated Monoclonal Antibody (FAB3832A) and either (A) Mouse Anti-Human EMR3 Monoclonal Antibody (Catalog # MAB8496) or (B) Mouse IgG2BFlow Cytometry Isotype Control (MAB0041) followed by Phycoerythrin-conjugated Anti-Mouse IgG Secondary Antibody (F0102B).
EMR3 in Human Lung Cancer Tissue. EMR3 was detected in immersion fixed paraffin-embedded sections of human lung cancer tissue using Mouse Anti-Human EMR3 Monoclonal Antibody (Catalog # MAB8496) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm in cancer cells. Staining was performed using our protocol for IHC Staining with VisUCyte HRP Polymer Detection Reagents.

Background: EMR3
EMR3 is a member of the class B seven-span transmembrane (TM7) receptor family. It is expressed predominantly by cells of the immune system, with highest levels in neutrophils, monocytes and macrophages. It may play a role in myeloid-myeloid interactions during immune and inflammatory responses. EMR3 upregulation in glioblastoma multiforme (GBM) is associated with poor survival and may be a potential therapeutic target.
