N-Acetyl-beta -D-Glucosa-minidase Release Induced by Complement Component C5a and Neutralization by Human Complement Component C5a R1 Antibody. Recombinant Human Complement Component C5a (Catalog # 2037-C5) induces N-acetyl-beta -D-glucosa-minidase release in the dibutyryl cyclic-AMP differentiated U937 human histiocytic lymphoma cell line in a dose-dependent manner (orange line). N-Acetyl-beta -D-Glucosaminidase Release elicited by Recombinant Human Complement Component C5a (10 ng/mL) is neutralized (green line) by increasing concentrations of Mouse Anti-Human Complement Component C5a R1 Monoclonal Antibody (Catalog # MAB3648). The ND50 is typically 2-10 µg/mL in the presence of cytochalasin-B.

Background: Complement Component C5aR1
Complement Component C5a R1 (C5aR1), also known as C5a ligand and CD88, is a 7TM protein expressed on myeloid, endothelial, epithelial, and smooth muscle cells. C5aR binds the activated complement anaphylatoxin C5a. In established allergic environments, this triggers neutrophil and eosinophil chemotaxis and the release of proinflammatory mediators. In contrast, C5aR1/C5a interactions are protective during allergen sensitization. Human C5aR1 shares 66% amino acid sequence identity with mouse and rat C5aR.
