Biological Activity

SB 431542 is a potent and selective inhibitor of the transforming growth factor-β (TGF-β) type I receptor/ALK5 (IC50 = 94 nM), and its relatives ALK4 and ALK7. Suppresses TGF-β-induced proliferation of human osteosarcoma cells. Replaces SOX2 in reprogramming of fibroblasts into iPSCs. Stimulates proliferation, differentiation and sheet formation of ESC-derived endothelial cells. Inhibits TGF-β-induced EMT, migration, invasion and VEGF secretion in several human cancer cell lines. Also used in a protocol to generate brain organoids from human iPSCs.

Tested a human lung epithelial cell line pretreated with SB431542 at either 10, 1 or 0.1uM for 30 min and then added TGFb1 at 5ng/ml in two different matrices. Incubated 24hr and then lysed cells to look at pSmad 2/3 levels by immunoassay
