Product Summary

Simple Plex assay for the detection of human CD14 in serum, plasma (EDTA/Heparin), and urine

Precision

Intra-Assay Precision (Precision within an assay) Each control was tested 16 times in one assay
Inter-Assay Precision (Precision between assays) Replicates of each control were tested in multiple assays performed by at least three technicians using two lots of reagents

Serum, EDTA Plasma, Heparin Plasma, Urine

Intra-Assay Precision Inter-Assay Precision
Sample 1 2 1 2
n 16 16 31 31
Mean (pg/mL) 31.7 1308 28.1 1233
Standard Deviation 3.07 95.9 2.74 162
CV% 9.7 7.3 9.7 13.2

Recovery

Recovery was not necessary as CD14 shows natural linearity data from a neat sample to a 1:16 dilution.

Linearity

Samples containing and/or spiked with high concentrations of human CD14 were serially diluted with Sample Diluent to produce samples within the dynamic range of the assay

Background: CD14

CD14 is an acute phase glycoprotein that binds lipopolysaccharide endotoxins (LPS) from Gram-negative bacteria. It is primarily expressed on monocytes, macrophages, and neutrophils, and soluble forms can be secreted or shed from the cell surface. CD14 accepts LPS from its initial binding by LBP, and it can further transfer LPS to the TLR4/MD2 complex. LPS acctivation of TLR4 triggers inflammatory activation. CD14 also contributes to the response to TLR3 ligands, phosphoinositides, as well as the surfactant proteins SP-A and SP-D.
