Simple Plex Control for Human Granzyme B Summary

INTENDED USE AND DESCRIPTION: For use a quantitative control for the determination of human and NHP Granzyme B concentrations in biological fluids. Controls are supplied lyophilized. Please refer to the lot specific certificate of analysis and Simple Plex Lyophilized Quality Control product insert for additional information.

Storage

• Store lyophilized controls at 2-8 °C. Lyophilized controls are stable for up to 12 months from date of receipt.
• Reconstituted controls are stable for up to 6 months from date of preparation when stored at ≤ -70 °C. Storage at -20° C is not recommended.
• Individual frozen aliquots of low and high controls are designed as single-use entities. Thaw directly before use and discard any remaining volume.

Background: Granzyme B

Granzyme family serine proteases are stored in the granules of cytotoxic T lymphocytes and natural killer cells. Granzymes contain one S1 protease domain and are synthesized as preproproteins. Granzymes are released toward pathogen infected or transformed target cells during cellular immune reactions. Target cell entry by granzymes is through perforin channels. Granzymes trigger apoptosis by caspase-dependent and -independent mechanisms. There are five granzymes (A, B, G, H and K) in human and many more in mouse and rat.
