Neutralization Antibody Validation and Quality

Tested in Relevant Bioassays

By utilizing our selection of in-house manufactured proteins, we are able to leverage our bioassay expertise to ensure that our antibodies block/neutralize the appropriate cellular function. Our assays include proliferation, chemotaxis, protein secretion, adhesion, and in vitro enzyme neutralization. We continuously add new assays to ensure the highest specificity based on the latest research data.

Low Endotoxin Levels

Our neutralizing antibodies are manufactured to ensure low endotoxin levels. Each lot is QC tested for the level of endotoxin and the information is reported on each individual datasheet. You won’t have to worry about endotoxin-induced nonspecific cellular responses.

What Is The Difference Between Blocking And Neutralizing Antibodies?

A blocking antibody binds its target and directly interferes with its function, such as blocking cell adhesion or receptor-ligand binding. A neutralizing antibody binds its target and negates its downstream cellular effects, such as cell proliferation or chemotaxis.

Cell Adhesion Mediated by Osteopontin/OPN and Neutralization by Human Osteopontin/OPN Antibody. Recombinant Human Osteopontin/OPN (Catalog # 1433-OP), immobilized onto a microplate, supports the adhesion of the HEK293 human embryonic kidney cell line in a dose-dependent manner (orange line). Adhesion elicited by Recombinant Human Osteopontin/OPN (1 µg/mL) is neutralized (green line) by increasing concentrations of Rabbit Anti-Human Osteopontin/OPN Monoclonal Antibody (Catalog # MAB14334). The ND50 is typically 0.5-1.5 µg/mL.

OPN blocking antibody blocks recombinant OPN in an adhesion assay.

Chemotaxis Induced by CXCL13/BLC/BCA-1 and Neutralization by Human CXCL13/BLC/BCA-1 Antibody. Recombinant Human CXCL13/BLC/BCA-1 (Catalog # 801-CX) chemoattracts the BaF3 mouse pro-B cell line transfected with human CXCR5 in a dose-dependent manner (orange line). The amount of cells that migrated through to the lower chemotaxis chamber was measured by Resazurin (Catalog # AR002). Chemotaxis elicited by Recombinant Human CXCL13/BLC/BCA-1 (0.05 µg/mL) is neutralized (green line) by increasing concentrations of Mouse Anti-Human CXCL13/BLC/BCA-1 Monoclonal Antibody (Catalog # MAB8012). The ND50is typically 0.5-2.5 µg/mL.
