Immunohistochemistry Survivin antibody in Human Breast Cancer Tissue by Immunohistochemistry (IHC-P). View Larger

Survivin in Human Breast Cancer Tissue. Survivin was detected in immersion fixed paraffin-embedded sections of human breast cancer tissue using Goat Anti-Human Survivin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6471) at 1 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Goat IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC004). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm. View our protocol for IHC Staining with VisUCyte HRP Polymer Detection Reagents.

Western Blot Detection of Human Survivin antibody by Western Blot. View Larger

Detection of Human Survivin by Western Blot. Western blot shows lysates of MCF-7 human breast cancer cell line and Jurkat human acute T cell leukemia cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human Survivin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6471) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for Survivin at approximately 18 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Simple Western Detection of Human Survivin antibody by Simple WesternTM. View Larger

Detection of Human Survivin by Simple WesternTM. Simple Western lane view shows lysates of Jurkat human acute T cell leukemia cell line, loaded at 0.2 mg/mL. A specific band was detected for Survivin at approximately 24 kDa (as indicated) using 10 µg/mL of Goat Anti-Human Survivin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6471) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Background: Survivin

Survivin also known as Apoptosis inhibitor survivin and Baculoviral IAP repeat-containing protein 5 (BIRC5) is an 18 kDa member of the inhibitor of apoptosis (IAP) gene family, which encodes negative regulatory proteins that prevent apoptotic cell death. IAP family members usually contain multiple Baculovirus IAP repeat (BIR) domains, but this gene encodes seven isoforms with only a single BIR domain. The encoded proteins also lack a C-terminus RING finger domain. Gene expression is high during fetal development and in most tumors, yet low in adult tissues. Survivin is expressed in a cell cycle-dependent manner and associates with the mitotic apparatus.

Human Survivin Antibody
By Abby Tyler on 11/07/2017
Application: ELISA Sample Tested: Serum and Plasma Species: Human

The antibody MAB886 was used as the capture in a sandwich ELISA along with AF6471 as detection. The ELISA worked very well. Healthy human serum and plasma samples were not detectable although some disease state samples are detectable.
