This DuoSet IC ELISA contains the basic components required for the development of sandwich ELISAs to measure phosphorylated human Chk2 (T68) in cell lysates. An immobilized capture antibody specific for Chk2 binds both phosphorylated and unphosphorylated Chk2. After washing away unbound material, a biotinylated detection antibody is used to detect only phosphorylated receptor, utilizing a standard HRP format.

Background: Chk2

The Chk2 checkpoint kinase is an integral member of the signaling cascade that controls cell cycle checkpoints. In response to genotoxic insults Chk2 is phosphorylated by ATM or ATM-related kinases. Phosphorylation of Chk2 T68 promotes the formation of Chk2 oligomers. Oligomerization is a critical event in the activation of the Chk2 kinase as it is required for phosphorylation of additional Chk2 residues that lead to full activation and signaling to downstream effectors.
