Scientific Data
Detection of Orexin R2/HCRTR2 in HEK293 Human Cell Line Transfected with Human Orexin R2/HCRTR2 and eGFP by Flow Cytometry. HEK293 human embryonic kidney cell line transfected with human Orexin R2/HCRTR2 and eGFP was stained with either (A) Mouse Anti-Human Orexin R2/HCRTR2 Monoclonal Antibody (Catalog # MAB5246) or (B) Mouse IgG2AIsotype Control (Catalog # MAB003) followed by Allophycocyanin-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # F0101B). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Detection of Orexin R2/HCRTR2 in A172 Human Cell Line by Flow Cytometry. A172 human glioblastoma cell line was stained with Mouse Anti-Human Orexin R2/HCRTR2 Monoclonal Antibody (Catalog # MAB5246, filled histogram) or isotype control antibody (Catalog # MAB003, open histogram). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Background: Orexin R2/HCRTR2
Hypocretin receptor 2 (HCRTR2); also known as orexin receptor 2 or OX2R is a 40 kDa 7-transmembrane G-protein-coupled glycoprotein that is a high affinity receptor for orexins A and B (hypocretins 1 and 2). In mouse brain, engagement of HCRTRs promotes wakefulness, such that absence of either orexins or their receptors creates a narcolepsy-like state. It also influences reward circuits involving food or addictive drugs. The extracellular portions of human HCRTR2 share 92% and 93% aa identity with corresponding portions of mouse and rat HCRTR2, respectively.
