Detection of Human/Mouse ZIC3 by Western Blot. Western blot shows lysates of 293T human embryonic kidney cell line, Jurkat human acute T cell leukemia cell line, and NIH-3T3 mouse embryonic fibroblast cell line. PVDF membrane was probed with 1 µg/mL of Human/Mouse ZIC3 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5310) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for ZIC3 at approximately 56 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

ZIC3 in A172 Human Cell Line. ZIC3 was detected in immersion fixed A172 human glioblastoma cell line using Human/Mouse ZIC3 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5310) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Sheep IgG Secondary Antibody (red, upper panel; Catalog # NL010) and counterstained with DAPI (blue, lower panel). Background: ZIC3
ZIC3 (zinc-finger protein of the cerebellum 3) is a 50 kDa (predicted) member of the GLI C2H2-type Zn-finger protein family. It is expressed in cerebellum, and apparently plays a key role in left-right body axis formation. Human ZIC3 is 467 amino acids (aa) in length. It contains N‑terminal poly-Ala (aa 46‑55) and poly-His (aa 87‑97) segments, followed by five C2H2-type Zn-fingers (aa 253‑410). Within the zinc finger region, there is an overlapping NES (aa 290‑349) and two NLS's (aa 367‑382 and 403‑410). ZIC3 reportedly binds to GLI-3. There is one potential splice form that shows a 49 aa substitution for aa 409‑467, and a second splice form that shows a three aa addition to the C‑terminus. Over aa 380‑467, human ZIC3 displays absolute/100% identity to mouse ZIC3.
