Detection of Human/Mouse AMPK beta 2 by Western Blot. Western blot shows lysates of A549 human lung carcinoma cell line and MRC-5 human embryonic lung fibroblast cell line. PVDF membrane was probed with 2 µg/mL Rat Anti-Human/Mouse AMPK beta 2 Monoclonal Antibody (Catalog # MAB3808) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). For additional reference, recombinant human AMPK beta 2 (0.5 ng) was included. A specific band for AMPK beta 2 was detected at approximately 34 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 3.

Background: AMPK beta 2
AMP-activated protein kinase (AMPK) is a heterotrimeric complex consisting of a catalytic alpha subunit and regulatory beta and gamma subunits. Each subunit exists as alternate isoforms ( alpha 1, alpha 2, beta 1, beta 2, gamma 1, gamma 2, gamma 3), with all 12 combinations able to form complexes. The beta 1 subunit is expressed at higher levels than the beta 2subunit in liver, while beta 2 is more abundant in skeletal muscle. AMPK’s role in metabolic regulation has implicated this serine/threonine kinase as a therapeutic target in heart disease, obesity, and diabetes.
