Carrier Free

What does CF mean?

CF stands for Carrier Free (CF). We typically add Bovine Serum Albumin (BSA) as a carrier protein to our recombinant proteins. Adding a carrier protein enhances protein stability, increases shelf-life, and allows the recombinant protein to be stored at a more dilute concentration. The carrier free version does not contain BSA.

What formulation is right for me?

In general, we advise purchasing the recombinant protein with BSA for use in cell or tissue culture, or as an ELISA standard. In contrast, the carrier free protein is recommended for applications, in which the presence of BSA could interfere.

Background: Fibronectin

Fibronectin (Fn) is a large modular glycoprotein that is found as a polymeric fibrillar network in the extracellular matirx (ECM) and as soluble disulfide-linked dimeric protomers in plasma and other body fluids. The protein subunit is made up of three types of homologous structural repeats termed Fn type I, type II, and type III repeats. Multiple isoforms of the protein formed by alternative splicing at numerous sites, resulting in insertions of extra type III domains (EDA and ECB) or parts of the variable type III connecting segment (V/IIICS), have been identified. Fibronectin is a ligand for fibrin, heparin, chondroitin sulfate, collagen/gelatin, and many integrin receptors. It is involved in multiple cellular processes including cell adhesion/migration, blood clotting, morphogenesis, tissue repair, and cell signaling. Fibronectin functions are mediated by the insoluble polymeric fibrils in the ECM. Conversion of the non-functional soluble Fibronectin to Fibronectin fibrils in the ECM is tightly regulated. The process is initiated by binding of Fibronectin to cell surface integrins, resulting in conformational changes and exposure of cryptic epitopes necessary for polymerization (1, 2).

FAQs

  1. What is the difference between Bovine Fibronectin Protein, CF (Catalog # 1030-FN) and Human Fibronectin Protein, CF (Catalog # 1918-FN)?

    • Only the source species for the protein is different. Both carrier-free proteins have been validated to be bioactive with the same assay protocols listed and recommended concentrations fall within the same range, although customers should determine their optimal concentrations for their particular cell type and application.

  2. What is the molecular weight of Human Fibronectin Protein, CF (Catalog # 1918-FN)?

    • This Human Fibronectin Protein is purified from human plasma. There are multiple isoforms of fibronectin that vary in molecular weight. Due to the varied weight of the different isoforms, the MW of purified fibronectin can run between 180-285 kDa in an SDS-PAGE gel under reducing conditions.

  3. The storage recommendation listed on the CoA and datasheet for Human Fibronectin Protein, CF (catalog # 1918-FN) says to store in a manual defrost freezer, yet store at 2 to 8 °C. Can 1918-FN be frozen?

    • Catalog #1918-FN should be stored exclusively at 2 to 8 °C and never frozen. We are working to correct the language about the manual defrost freezer.
