Detection of DC‑LAMP in Human Mature Dendritic Cells by Flow Cytometry. Human mature dentritic cells differentated from human peripheral blood mononuclear cell derived CD14+cells treated with 20 ng/mL Recombinant Human IL-4 (204-IL) and 50 ng/mL Recombinant Human GM-CSF (215-GM) for 7 days and 1 µg/mL LPS, 20 ng/mL Recombinant Human TNF-a (210-TA), and 20 ng/mL Recombinant Human IL-1 beta /IL-1F2 (201-LB) for last 24 hours were stained with Goat Anti-Human DC-LAMP Lumenal Domain Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4087, filled histogram) or isotype control (AB-108-C, open histogram), followed by Donkey Anti-Goat IgG (H+L) Allophycocyanin (F0108).

Background: DC-LAMP
DC-LAMP (dendritic cell lysosome-associated membrane protein: also known as CD208 and LAMP3) is a 50‑70 kDa member of the LAMP family of proteins. Mature human DC-LAMP is a 389 amino acid (aa) type I transmembrane protein. It has a 254 aa lumenal N‑terminus and a short 14 aa cytoplasmic tail. The molecule is found in type II pneumocytes, interdigitating DC, and various tumors. DC-LAMP serves as a marker of human mature DC, and it may play a role in normal lysosome and endosome function. Over the region used as immunogen, human DC-LAMP shares 72% and 55% aa sequence identity with canine and mouse DC-LAMP, respectively.
