Why Use ExCellerate™ iPSC Expansion Medium?
ExCellerate™ iPSC Expansion Medium, Animal-Free, GMP is specifically formulated for the in vitro, feeder-free expansion of human pluripotent stem cells, including embryonic stem (ES) and induced pluripotent stem (iPS) cells. The formulation is complete and contains the growth factors required to support iPSC expansion and maintenance in a single, ready-to-use format. This simplifies stem cell culture workflows and provides a reproducible cell culture environment to ensure consistent performance.
No materials of animal or human origin are used in the production of this medium or its components. This medium is produced in a dedicated Animal-Free production space. Elimination of animal-derived components reduces variability and improves lot-to-lot consistency for reliable generation of high quality, homogenous, and undifferentiated cells needed for any stage of research. Animal-free traceability of raw materials reduces risk and makes it ideal for translational research. An RUO version is also available to support a seamless transition into clinical applications.
The composition of ExCellerate™ iPSC Expansion Medium is optimized to support the robust expansion of pluripotent stem cells, while maintaining cell viability, genotype and differentiation potential for extended times in vitro. WiCell has concluded that ExCellerate™ iPSC Expansion Medium has met all WiCell requirements for quality and is appropriate for pluripotent stem cell culture.
This medium is compatible with various cell culture matrices, including Cultrex Ultimatrix, Cultrex ReadyBME and Recombinant Human Vitronectin, and cell lines, making it versatile for use in research, preclinical, and translational stem cell culture workflows.

Human iPSC expanded in CCM036 and CCM036-GMP show similar growth kinetics. Catalog # CCM036 (RUO version) and Catalog # CCM036-GMP have the same formulation and are manufactured with the same high quality animal-free components. No significant difference in growth kinetics is seen when culturing iPSC in either CCM036 or CCM036-GMP.
Using ExCellerate iPSC Media better supports low density iPSC culture compared to other media suppliers. Human iPSCs were plated at low density (3000 cells per well in a 6 well plate) and cultured until the cells reached approximately 70% confluence for 3 passages. Cells were harvested using Accutase and live cells were counted. All groups were then normalized to CCM036-GMP. Competitors showed slower and highly variable cell growth (Plot A) as well as lower viability (Plot B) compared to CCM036-GMP.
ExCellerate™ iPSC Expansion Medium maintain expression of stemness markers over long-term cultures (at least 24-passages). Human iPSC expanded in CCM036-GMP express key stemness markers, Oct 3/4 and Nanog. SSEA-1 used as negative control.
Stable genotype over long-term culture. Human iPSCs cultured in CCM036-GMP show characteristics healthy colony morphology including clearly defined edges, compact and tightly packed cells, along with normal karyotype after 26 passages.

iPSC grown in CCM036-GMP retain their pluripotency. After expansion in CCM036-GMP, cells were differentiated to the three germ layers: endoderm (SOX17), mesoderm (Brachyury), and ectoderm (OTX2).
