Ready-to-use, non-enzymatic solution to depolymerize basement membrane matrix for harvesting organoids from culture.

Why Use Cultrex Organoid Harvesting Solution?

Organoid cultures exhibit cellular behaviors and morphologies similar to those seen in vivo, however, the adaptation of these models for studying biochemical processes has been impeded by the challenge of separating intact organoids from extra-cellular proteins comprising the hydrogel. Commonly, proteases are employed to degrade these extracellular proteins, however, proteases also degrade proteins on the cell surface and protease activity may carry over into subsequent cultures or lysate preparations. Cultrex Organoid Harvesting Solution provides a non-enzymatic method for depolymerizing extracellular matrix proteins to allow for harvesting of intact organoids for passaging, cryopreservation, or biochemical analysis.

Negative for the presence of bacteria and fungi.

    • The Organoid Harvesting Solution works well with BME (Basement Membrane Extract).  For cells grown on Cultrex Rat Collagen 1, the recommended enzyme to use to dissociate cells is Collagenase. Collagenase treatment at 37oC for 30 minutes is recommended.

  2. Are Cultrex Organoid Harvesting Solution (Catalog # 3700-100-01) and dispase interchangeable?

    • Cultrex Organoid Harvesting Solution (Catalog # 3700-100-01) is a non-enzymatic solution that depolymerizes the matrix to enable harvest of organoids. In contrast, dispase is a protease enzyme that degrades the matrix. End users will need to empirically determine whether enzymatic or non-enzymatic solutions are more appropriate for their experimental setup.

I have been using Organoid Harvesting Solutions since 2018 for organoid models biochemical analysis.
It's very easy to use and allows you to dissociate and depolymerize any basement membrane matrix (Ultimatrix, BME Type 2 Selec, Matrigel) to harvest intact organoids.
